Zafirlukast works by blocking the action of certain natural substances that cause swelling and tightening of the airways. Specifically, Zafirlukast acts as a selective and competitive antagonist of leukotriene D4 (LTD4) and leukotriene E4 (LTE4), which are components of the slow-reacting substance of anaphylaxis (SRSA). These cysteinyl leukotrienes play an important role in the pathophysiology of asthma by promoting airway edema, smooth muscle constriction, and inflammatory cellular activity, all of which contribute to the signs and symptoms of asthma.

Zafirlukast is commonly available as 10 mg and 20 mg oral tablets and is typically taken twice daily, either 1 hour before or 2 hours after meals.
